非离子型UV固化聚氨酯水凝胶的合成与性能研究

摘要:以聚乙二醇2000(PEG2000)、异佛尔酮二异氰酸酯(IPDI)、1,4丁二醇(BDO)、丙三醇(GLY)、丙烯酸2羟乙酯(HEA)为主要原料,采用丙酮预聚体法合成

了一系列透明的、非离子型UV固化聚氨酯水凝胶.研究了BDO/GLY的配比对水凝胶溶胀性能、拉伸性能的影响;探讨了温度、离子强度对水凝胶溶胀性能的影响;并以硫酸庆大霉素作为模拟药物,对其在模拟体液PBS缓冲溶液中的释放进行了初步探讨.当nBDO/nGLY为1

时,水凝胶干膜拉伸强度达到1.46 MPa,伸长率为87%,20 ℃去离子水中溶胀比达到370%,且表现出显著的温度敏感性和较好的离子强度稳定性,并具有载药和释放药物的能力.

Abstract:A series of transparent UVcurable nonionic polyurethane (PU) hydrogel were prepared by the prepolymerization of polyethylene glycol 2000(PEG2000), isophorone diisocyanate (IPDI), 2hydroxyethyl acrylate (HEA), 1,4butanediol (BDO) and glycerol (GLY). The influence of the proportion of BDO to GLY on swelling properties and tensile properties of PU hydrogel was studied. The effects of temperature and ionic strength on the swelling ratio (SR) of PU hydrogel were also investigated. The results indicated that when

nBDO/nGLY=1, the tensile strength of PU hydrogel dry film reached 1.46 MPa, elongation was 87%, and SR was 370% in the deionized water at 20 ℃. Meanwhile, PU hydrogel showed strong temperature sensitivity and good stability to ionic strength. The release of the gentamycin sulfate from the swelling hydrogel in PBS buffer solution was eventually investigated to verify whether the obtained hydrogel can be used in drug delivery.

水凝胶是一种由具有亲水性聚合物交联网络与其吸附溶胀的水构成的特殊分散体系[1-2].

它能够在水中溶胀而不溶解,由于其含有大量的水类似于细胞外基质,对体液及人体组织表现出很好的生物相容性,可作为药物缓释载体、组织填充材料、创面敷料等,因而在生物医药领域得到了广泛的研究与应用[3-4].特别是在伤口敷料方面,水凝胶敷料可防止伤口过于湿润或

干燥,能够吸收伤口表面的渗出物,促进伤口愈合.此外水凝胶敷料还能够阻止新形成的肉芽

组织黏附在敷料表面从而避免在摘除敷料时带来的二次痛苦[5-6].
