酸-热处理法破碎酵母细胞的研究

摘要:　本试验研究了酸-热处理法进行酵母细胞破碎时,其酸浓度大小、热处理时间、浸泡时间、酸用量对破碎效果的影响。正交试验结果表明,破碎条件的最佳组合为HC l 浓度1mol/l,HCl 用量12m l/g 干细胞,浸泡时间60m in,热处理时间4m i n 。

胞壁破碎是提取胞内产物的关键步骤[4]。在酵

母菌体蛋白含量的测定过程中,细胞壁破碎方法的

选择相当重要。目前,细胞破碎的方法很多,可分为

机械法和非机械法。机械法(如珠磨法、高压匀浆

法、超声波法)破碎细胞是用高速、研磨或超声波空

化现象等造成剪切、碰撞,从而造成细胞壁和膜被撕

裂、破碎[1]。机械法的最大缺点在于所得处理液粘度很高,难以进行离心分离,且由于菌体破坏后形成许多杂蛋白,影响了后续的分离测定。非机械法包括溶酶法、渗透压冲击、冻融法、干燥法、化学渗透法等。溶酶法是研究较广的1种方法,它利用酶反应,分解破坏细胞壁上特殊的键,从而达到破壁的目的。常用的溶酶有溶菌酶、β-1,3-葡聚糖酶、β-1,6-葡聚糖酶、甘露糖酶、糖苷酶、肽链内切酶、壳多糖酶
