小檗碱对胃癌SGC7901细胞增殖和凋亡的影响

小檗碱对胃癌SGC7901细胞增殖和凋亡的影响

摘要:目的:探讨小檗碱对胃癌细胞系SGC7901的抑制作用及相关机制。方法:采用MTT的方法观察不同

浓度小檗碱对SGC7901细胞的24h和48h抑制率,并计算24h和48h的50%抑制浓度(IC 50)值;采用流式细胞技术

检测不同浓度小檗碱作用48h后胃癌细胞系SGC7901凋亡的发生率和细胞周期的变化。结果:小檗碱对胃癌细胞

SGC7901生长的抑制作用呈现出时间和剂量依赖性。24h和48h IC 50分别为74.16、36.84μmol/L。1μmol/L至120μmol/L

浓度梯度的小檗碱干预48h后,SGC7901凋亡率发生率逐渐增加。其中5、10、25、50、75、120μmol/L与对照组

比较有统计学差异(P <0.05),120μmol/L时凋亡发生率达到35.43%;5、25、75μmol/L浓度的小檗碱作用后,

SGC7901胃癌细胞G 0/G 1期细胞比例较对照组明显升高(P <0.05),而S期细胞比例则随浓度增加呈现出下降趋势,

其中25、75μmol/L浓度组与对照组比较,差异有统计学意义(P <0.05),对G 2/M期细胞,各组比例变化无明显差

别。结论:小檗碱对胃癌细胞系SGC7901具有时间和浓度依赖性的抑制作用,抑制作用与阻滞细胞周期于G 0/G 1

期,抑制DNA合成和诱导凋亡有关。

Abstract: Objective: To investigate the effect of berberine on inhibition of gastric adenocarcinoma cell lines SGC7901

and relative mechanisms. Methods: MTT was conducted to essay inhibition rates at different concentrations of berberine on

SGC7901 cells in 24h and 48h, and the half maximal inhibitory concentration (IC 50) was calculated according to inhibition

rates. Apoptosis ratios and cell cycle changes in SGC7901 were determined by ? ow cytometry after 48h treating with different

concentration berberine. Results: Berberine embraced the time and dose dependent activity of growth inhibition in gastric cancer

cells. IC 50 at 24 and 48 hours were 74.16μmol/L and 36.84μmol/L respectively. In addition, apoptosis ratio in SGC7901 indicated

an gradually increasing trend with concentration gradient (1μmol/L to 120μmol/L) after 48h intervention of berberine, and there

were statistically signi ? cant differences (P <0.05) in 5, 10, 25, 50, 75, 120μmol/L groups compared with the control group, the rate

of apoptosis went up to the climax at 35.43% in 120μmol/L group. For cell cycle, the proportion of SGC7901 in G 0/G 1 phase in

5, 25, 75μmol/L group respectively after 48h treatment of berberine with statistically signi ? cant difference in comparison to the

control group (P <0.05). In contrast, the proportion in S phase showed a downward tendency with the increasing concentration,

and there was statistical decreasement in 25, 75μmol/L group (P <0.05) compared to the control group. Moreover no statistical

difference was observed in aspect of the G 2/M phase among the groups. Conclusion: Berberine inhibited the proliferation of gastric

cancer cell SGC7901 in a time and concentration dependent manner associated with cycle arrest in G 0/G 1 phase, inhibition of DNA

synthesis and induction of apoptosis.
